In the study described below, scientists examined the metabolic profiles of drostanolone in urine using tandem quadrupole time-of-flight mass spectrometry (LC-QTOF-MS) liquid chromatography methods in full scan mode and targeted detection of MS / MS with accurate mass measurement.  

The study involved one healthy male volunteer who had been injected with masteron steroid in USA, and liquid-liquid extraction was used along with direct injection analysis to analyze urine samples. Chromatographic peaks of potential metabolites were identified with an estimated [MH] (-) as the target ion in the full scan study, and deprotonated ions were analyzed in the MS / MS target mode.

As a result, it was confirmed that the presence of drostanolone ( masteron) indicate eleven metabolites: five new sulfates, five glucuronide conjugates and one free metabolite. Due to the lack of fragment ions suitable for determining the structure of steroid rings of phase II drostonalone metabolites, gas chromatography mass spectrometry (GC-MS) was used to determine the exact structure of the metabolite of trimethylsilylated phase I released after enzymatic hydrolysis, and the potential structure was determined using combined mass spectrometry method. For metabolites, the time of eventual detection was recorded. S4 (2?-methyl-5?-androstan-17-one-6?-ol-3?-sulfate) and G1 (2?-methyl-5?-androstan-17-one-3?-glucuronide) can be considered new potential biological markers of drostanolone. They can be detected in urine.within 24 days using the liquid-liquid extraction method and within 7 days by direct injection analysis after intramuscular injection. S4 and G1 were also found in two urine samples in which drostanolone was known to be present.
